PE-22-28 is a synthetic seven-amino-acid peptide developed in 2017 by Djillani and colleagues as a shortened, optimized analogue of spadin, a naturally occurring peptide cleaved from the propeptide of the sortilin/neurotensin-receptor-3 precursor. Compared with the parent spadin, PE-22-28 was engineered for markedly greater potency and improved in-vivo stability, with reported TREK-1 inhibition in the sub-nanomolar range and a substantially extended duration of action in animal studies. This makes it a valued laboratory tool for probing two-pore-domain potassium-channel biology. Within neuropharmacology research it is distinguished as one of the most potent spadin-derived TREK-1 blockers characterized to date in preclinical behavioral and cellular models.
Mechanism & Research Context
PE-22-28 is studied as a selective blocker of TREK-1 (TWIK-related potassium channel 1), a two-pore-domain background potassium channel expressed in brain regions implicated in mood regulation. Because TREK-1-knockout animals display a depression-resistant phenotype, the channel is a validated research target. In rodent models PE-22-28 inhibition of TREK-1 is investigated for downstream effects on neuronal excitability, hippocampal neurogenesis, and antidepressant-like behavioral endpoints observed within days of administration.
