SNAP-8, known by its INCI name acetyl octapeptide-3, is a synthetic acetylated octapeptide with the sequence Ac-Glu-Glu-Met-Gln-Arg-Arg-Ala-Asp and a molecular weight near 1076 Da. It is modeled on the N-terminal region of SNAP-25, the synaptosomal-associated protein that contributes two of the four helices forming the neuronal SNARE complex. An extended analogue of the hexapeptide argireline, SNAP-8 is distinguished by its longer sequence and by acting as a mimetic that competes for SNARE assembly sites. This makes it a useful tool compound for studying SNARE-dependent vesicle fusion and neurotransmitter-release mechanisms in cell-culture and biochemical research.
Mechanism & Research Context
SNAP-8 presents a sequence mirroring the SNAP-25 N-terminus and is studied as a competitive interferer with SNARE four-helix-bundle assembly, disrupting alignment of syntaxin, synaptobrevin, and SNAP-25 helices. Unlike botulinum toxin’s irreversible proteolytic cleavage, this competitive, reversible mechanism reduces rather than abolishes vesicle-fusion efficiency. In vitro, it has been reported to lower stimulated neurotransmitter release, and it is used to probe exocytosis and SNARE-modulation pathways in neuronal and secretory-cell research models.
