MOTS-c is a 16-amino-acid mitochondrial-derived peptide encoded by a short open reading frame within the mitochondrial 12S ribosomal RNA gene, rather than by the nuclear genome. It was among the first mitochondrial peptides shown to act as a signaling molecule that influences nuclear gene expression and systemic metabolism, exemplifying retrograde mitochondrial-to-nuclear communication. Its expression is reported to rise in response to metabolic stress and exercise, and under such conditions it can translocate to the nucleus. These features make MOTS-c a distinctive research tool for studying how mitochondria encode regulatory peptides that coordinate cellular energy metabolism, stress adaptation, and aging-related processes in cell and animal models.

In research models MOTS-c is studied for its action on the folate-methionine cycle and de novo purine biosynthesis, where inhibition promotes accumulation of AICAR, an activator of AMP-activated protein kinase (AMPK). AMPK activation is examined for downstream effects on energy homeostasis, insulin sensitivity, and inflammatory signaling. Under stress, nuclear-translocated MOTS-c is also studied for binding antioxidant-response-element regulatory regions and modulating stress-adaptation gene programs alongside SIRT1 and PGC-1alpha pathways.
