Thymosin Alpha-1 (TA-1, thymalfasin) is a 28-amino-acid acetylated peptide originally isolated from thymic tissue as a fragment of the precursor prothymosin alpha. One of the most extensively studied immunomodulatory peptides, it is a defined single-sequence peptide rather than a crude thymic extract, and is approved in numerous countries as an immune adjuvant. Structurally acidic and highly conserved, it acts on innate and adaptive immune cells to shape cytokine output. It is distinguished from mixed thymic preparations such as thymalin and from the actin-binding beta-thymosins, occupying a distinct role in immune-signaling research.

Mechanism & Research Context

Thymosin Alpha-1 signals largely through Toll-like receptors, notably TLR-2 and TLR-9 on dendritic cells and other myeloid populations, engaging MyD88-dependent NF-kappaB and IRF signaling. This promotes dendritic-cell maturation, Th1-polarizing cytokines such as IL-12 and interferons, and enhanced T-cell and NK-cell activity in culture. It also induces indoleamine-2,3-dioxygenase in plasmacytoid dendritic cells, supporting regulatory T-cell balance. Preclinical models use it to study immune restoration, antiviral and antitumor responses, and cytokine modulation.
